Western blot analysis of Calcium Sensing Receptor/CASR using anti-Calcium Sensing Receptor/CASR antibody (PB9924). The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human 22RV1 whole cell lysates.
After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-Calcium Sensing Receptor/CASR antigen affinity purified polyclonal antibody (PB9924) at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for Calcium Sensing Receptor/CASR at approximately 130 kDa. The expected band size for Calcium Sensing Receptor/CASR is at 121 kDa.

Calcium Sensing Receptor/CASR was detected in a paraffin-embedded section of mouse kidney tissue. Biotinylated goat anti-rabbit IgG was used as secondary antibody. The tissue section was incubated with rabbit anti-Calcium Sensing Receptor/CASR Antibody (PB9924) at a dilution of 1:200 and developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB (Catalog # AR1027) as the chromogen.

The calcium-sensing receptor (CaSR) is a G protein-coupled receptor that is expressed in the parathyroid hormone (PTH)-producing chief cells of the parathyroid gland, and the cells lining the kidney tubule. It senses small changes in circulating calcium concentration and couples this information to intracellular signaling pathways that modify PTH secretion or renal cation handling, thus this protein plays an essential role in maintaining mineral ion homeostasis. Mutations in this gene cause familial hypocalciuric hypercalcemia, familial, isolated hypoparathyroidism, and neonatal severe primary hyperparathyroidism.
