Western blot analysis of anti-Collagen Type I/COL1A1 antibody (PB0980). The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human placenta tissue lysates,
Lane 2: rat skin tissue lysates,
Lane 3: rat lung tissue lysates,
Lane 4: mouse skin tissue lysates,
Lane 5: mouse NIH/3T3 whole cell lysates.
After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-Collagen Type I/COL1A1 antigen affinity purified polyclonal antibody (PB0980) and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for Collagen Type I/COL1A1 at approximately 130,220 kDa. The expected band size for Collagen Type I/COL1A1 is at 139 kDa.

Collagen, type I, alpha 1, also known as COL1A1, is a human gene that encodes the major component of type I collagen, the fibrillar collagen found in most connective tissues, including cartilage. This gene is mapped to 17q21.33. And this gene encodes the pro-alpha1 chains of type I collagen whose triple helix comprises two alpha1 chains and one alpha2 chain. Type I is a fibril-forming collagen found in most connective tissues and is abundant in bone, cornea, dermis and tendon. Mutations in this gene are associated with osteogenesis imperfecta types I-IV, Ehlers-Danlos syndrome type VIIA, Ehlers-Danlos syndrome Classical type, Caffey Disease and idiopathic osteoporosis.

注意事项

WB实验中，COL1A1在样本中存在很多的剪切位点，如果样本处理不当，很容易发生降解或是剪切，因此建议使用新鲜制备的样本，同时提高裂解液中蛋白酶抑制剂的浓度，最大程度避免蛋白发生降解。如果组织样本常规提取的蛋白检测不到或检测条带很弱，可以尝试用以下方式制备样本：
1. 取组织，剔去组织外部脂肪及结缔组织。
2. 丙酮中浸泡过夜，进一步去除残留脂肪，用冷蒸馏水清洗，除去丙酮（若组织本身不含脂肪，可以省略此步）。
3. 将组织切碎，倒入少量0.5mol/L乙酸（pH2.5），用打碎机打制成匀浆，加入胃蛋白酶。酶量与湿组织量之比为1:100（g/ml），再加入足量0.5mol/L乙酸，使溶液中酶的终浓度为2mg/ml。搅拌过夜（建议48小时）。16,000r、45min离心。
4. 留上清。沉淀部分重复第3步，弃去沉淀。将2次上清合并。
5. 上清边搅拌边缓缓加入氯化钠粉末，使终浓度至0.7mol/L（1 L溶液中含有40.95 g氯化钠），放置过夜。16,000r、45min离心，弃上清。
6. 沉淀用匀浆器匀浆，加0.5mol/L乙酸溶液，使溶液完全透明。
7. 边搅拌边缓慢加入氯化钠，使终浓度为0.7mol/L，放置过夜。
8. 16,000r×45min离心，弃上清。沉淀即为组织胶原。
9. 沉淀用2M尿素溶解，再加等体积的2X上样缓冲液混匀，置100℃水浴箱煮沸变性5分钟，至此样本制备完成。
