Western blot analysis of NG2/CSPG4 using anti-NG2/CSPG4 antibody (A03394-3). The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human A375 tissue lysates.
After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-NG2/CSPG4 antigen affinity purified polyclonal antibody (A03394-3) at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for NG2/CSPG4 at approximately 280,310 kDa. The expected band size for NG2/CSPG4 is at 251 kDa.

Chondroitin sulfate proteoglycan 4, also known as melanoma-associated chondroitin sulfate proteoglycan (MCSP) or neuron-glial antigen 2 (NG2), is a chondroitin sulfate proteoglycan that in humans is encoded by the CSPG4 gene. CSPG4 plays a role in stabilizing cell-substratum interactions during early events of melanoma cell spreading on endothelial basement membranes. It represents an integral membrane chondroitin sulfate proteoglycan expressed by human malignant melanoma cells.
