Western blot analysis of DRP1/DNM1L using anti-DRP1/DNM1L antibody (A00556-3). The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: HepG2 whole cell lysates,
Lane 2: Hela whole cell lysates,
Lane 3: A549 whole cell lysates,
Lane 4: rat heart tissue lysates.
After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-DRP1/DNM1L antigen affinity purified polyclonal antibody (A00556-3) at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for DRP1/DNM1L at approximately 78-82 kDa. The expected band size for DRP1/DNM1L is at 82 kDa.

背景资料

Dynamin-1-like protein is a GTPase that regulates mitochondrial fission. In humans, dynamin-1-like protein, which is typically referred to as dynamin-related protein 1 (Drp1), is encoded by the DNM1L gene. The encoded protein mediates mitochondrial and peroxisomal division, and is involved in developmentally regulated apoptosis and programmed necrosis. Dysfunction of this gene is implicated in several neurological disorders, including Alzheimer's disease. Mutations in this gene are associated with the autosomal dominant disorder, encephalopathy, lethal, due to defective mitochondrial and peroxisomal fission (EMPF). Alternative splicing results in multiple transcript variants encoding different isoforms.
