Western blot analysis of Endostatin/COL18A1 using anti-Endostatin/COL18A1 antibody (A01302-1). The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human COLO-320 whole cell lysates,
Lane 2: human placenta tissue lysates.
After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-Endostatin/COL18A1 antigen affinity purified polyclonal antibody (A01302-1) at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for Endostatin/COL18A1 at approximately 180 kDa. The expected band size for Endostatin/COL18A1 is at 178 kDa.

Collagen alpha-1(XVIII) chain is a protein that in humans is encoded by the COL18A1 gene. This gene encodes the alpha chain of type XVIII collagen. This collagen is one of the multiplexins, extracellular matrix proteins that contain multiple triple-helix domains (collagenous domains) interrupted by non-collagenous domains. A long isoform of the protein has an N-terminal domain that is homologous to the extracellular part of frizzled receptors. Proteolytic processing at several endogenous cleavage sites in the C-terminal domain results in production of endostatin, a potent antiangiogenic protein that is able to inhibit angiogenesis and tumor growth. Mutations in this gene are associated with Knobloch syndrome. The main features of this syndrome involve retinal abnormalities, so type XVIII collagen may play an important role in retinal structure and in neural tube closure. Alternative splicing results in multiple transcript variants.
