Western blot analysis of anti- PLEC antibody (PB9430). The sample well of each lane was loaded with 30ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates.
Use rabbit anti- PLEC 1:1000, probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog#EK1002). A specific band was detected for PLEC at approximately 532KD. The expected band size for PLEC is at 532KD.

Plectin/PLEC was detected in an immunocytochemical section of U2OS cells. The section was incubated with rabbit anti-Plectin/PLEC Antibody (PB9430) at a dilution of 1:100. Fluoro488 Conjugated Goat Anti-Rabbit IgG (Green) (Catalog # BA1127) was used as secondary antibody. The section was counterstained with DAPI (Catalog # AR1176) (Blue).

Plectin, known as PLEC, is a prominent member of an important family of structurally and in part functionally related proteins, termed plakins or cytolinkers, that are capable of interlinking different elements of the cytoskeleton. Plakins, with their multi-domain structure and enormous size, not only play crucial roles in maintaining cell and tissue integrity and orchestrating dynamic changes in cytoarchitecture and cell shape, but also serve as scaffolding platforms for the assembly, positioning, and regulation of signaling complexes. Plectin is expressed as several protein isoforms in a wide range of cell types and tissues from a single gene located on chromosome 8 in humans. The plectin gene locus in mouse on chromosome 15 has been analyzed in detail, revealing a genomic exon-intron organization with well over 40 exons spanning over 62 kb and an unusual 5' transcript complexity of plectin isoforms. Eleven exons (1-1j) have been identified that alternatively splice directly into a common exon 2 which is the first exon to encode plectin's highly conserved actin binding domain (ABD).
