一.凋亡酶caspase通路
细胞凋亡是一种受调节的细胞自杀机制,通常表现为核浓缩、起皱、膜发泡以及DNA片段化。Caspase家族属于半胱氨酸蛋白酶。起始组Caspase包括caspase-2,-8,-9,-1 0,-11和-12,与促凋亡信号紧密相连,一旦激活,这些酶会切割并激活下游的效应组Caspas e,包括Caspase-3,-6,-7,效应Caspase通过对细胞内蛋白特定的天冬氨酸残基位置处进行切割实现细胞的凋亡。FasL和TNF对Fas和TNFR的结合能够激活caspase-8和-10, DNA损伤诱导PIDD的表达, PIDD与RAIDD和caspase-2结合并激活caspase-2。

二.线粒体凋亡通路
Bcl-2家族通过控制线粒体膜的通透性来调控凋亡。抗凋亡蛋白Bcl-2和Bcl-xL定位于线粒体外膜,抑制细胞色素C的释放。促凋亡的Bcl-2家族成员Bad, Bid, Bax和Bim可能定位于胞浆,在收到凋亡信号后,转移到线粒体中,促进细胞色素C的释放。Bad转移到线粒体后与Bcl-xL形成促凋亡复合体。这个转移过程被生存因子所抑制,生存因子引起Bad的磷酸化而后在胞浆中富集。被Fas活化的caspase-8剪切胞浆Bid,所形成的有效分子tBid转移到线粒体上。 Bax和Bim在受到死亡刺激后转移到线粒体上,包括生存因子的消失。p53受DNA损伤的活化后能够诱导Bax, Noxa和Puma的转录。从线粒体中释放后,细胞色素C与Apaf-1结合并与caspase-9形成活性复合体。虽然在凋亡中线粒体通透性调节和细胞色素C释放的调节机制并没有完全阐明,但Bcl-xL, Bcl-2和Bax可能影响膜电位依赖的阴离子通道(VDAC, volta ge-dependent anion channel),这可能在调节细胞色素C中发挥作用。Mule/ARF-BP1是一个DNA损伤激活的E3泛素连接酶,它针对p53和Mcl-1 , Mcl-1是一个Bcl-2家族中的抗调亡分子。

三.死亡受体凋亡通路
死亡受体包括Fas, TNFaR, DR3, DR4和DR5,当配体与之结合后,这些蛋白被激活并起始凋亡。通常死亡配体通过使受体多聚化启动信号传导,受体多聚化后引起相关接头蛋白的结合并激活caspase级联。FasL结合Fas后形成Fas三聚体,通过接头蛋白FADD召集caspa se-8,然后caspase-8形成寡聚体通过自我剪切而活化。caspase-8通过两条平行的通路促进凋亡:它可以直接剪切并激活caspase-3也可以切割Bid,这是一个促凋亡的Bcl-2家族成员。截短的Bid(tBid,truncated Bid)转移进入线粒体,促进细胞色素C的释放,从而导致caspase-9和3的激活。TNF-a和DR-3L可以同时传递促凋亡和抑凋亡信号。TNFaR和DR3通过接头蛋白TRADD/FADD活化caspase-8促进凋亡.TNF-a与TNFaR结合后也能通过NIK/IKK激活 NF-KB通路。活化的NF-KB诱导促生存因子Bcl-2和FLIP的表达, FLIP直接抑制caspase-8的激活。FasL和TNF-a还可以通过ASK1/MKK7激活INK,活化的JNK可以磷酸化并抑制Bcl-2 。当级联通路未被激活时,死亡受体的活化可以通过形成复合1lb来激活另一个过程-细胞程序性死亡,又名坏死性凋亡。

四.凋亡抑制剂和凋亡促进剂
细胞生存需要积极的抑制凋亡发生,一方面需要抑制促凋亡因子的表达,另一方面则需要表达一些抗凋亡因子。PI3K通路被许多生存因子活化,能够激活Akt, Akt是生存信号传导中一个重要的角色。PTEN抑制PI3K通路。活化的Akt抑制促凋亡Bcl-2家族成员Bad, Bax, c aspase-9, GSK-3和Fox01,许多生长因子和细胞因子诱导抗凋亡Bcl-2家族成员。激酶Jaks和Src磷酸化并激活Stat3,后者诱导Bcl-xL和Bcl-2的表达。Erk1/2和PKC激活p9ORSK,后者激活CREB并诱导Bcl-xL和Bcl-2的表达。这些Bcl-2家族成员保护线粒体的完整性,阻止细胞色素C的释放以及随后的caspase-9的激活。TNF-a可以同时激活促凋亡和抑制凋亡通路: TN F-a活化caspase-8和-10诱导凋亡,同时它又通过NF-KB抑制凋亡, NF-KB诱导抗凋亡基因如 Bcl-2的表达。clAP1/2能与TRAF2结合抑制TNF- 的信号传导。FLIP抑制caspase-8的激活。

五.细胞程序化坏死
是指细胞坏死过程受精密信号控制,也叫坏死凋亡,是一种有别于凋亡的程序性细胞死亡方式。程序性坏死与传统的坏死特征类似,质膜破裂,引发炎症反应。其与缺血性损伤,神经退行性疾病有关,是近年来细胞死亡研究的热点。

六.细胞焦亡
又称细胞炎性坏死,是—种程序性细胞死亡,表现为细胞不断胀大直至细胞膜破裂,导致细胞内容物的释放进而激活强烈的炎症反应,是机体一种重要的天然免疫反应。其参与感染性疾病、神经系统相关疾病和动脉粥样硬化性疾病等的发生发展,并发挥重要作用。

七.细胞铁死亡
该概念最早由哥伦比亚大学的Brent Stoc kwell教授于2012年提出,是指—种由于细胞内代谢通路紊乱导致脂质过氧化物过度积累引起的细胞死亡方式,与细胞内的铁代谢和脂质稳态密切相关。
