Western blot analysis of anti-P-HSP27 antibody (BM4540). The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human MCF-7 whole cell lysates,
Lane 2: human Hela whole cell lysates,
Lane 3: rat PC-12 whole cell lysates,
Lane 4: rat C6 whole cell lysates,
Lane 5: mouse HEPA1-6 whole cell lysates,
Lane 6: mouse RAW264.7 whole cell lysates.
After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-P-HSP27 antigen affinity purified monoclonal antibody (BM4540) at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for P-HSP27 at approximately 27 kDa. The expected band size for P-HSP27 is at 23 kDa.

The heat-shock proteins(HSPs) belong to a larger group of polypeptides, the stress proteins, that are induced in various combinations in response to environmental challenges and developmental transitions. Heat-shock 27-kD protein1also knows as HSPB. Synthesis of the small(27-kD) HSP has been shown to be correlated with the acquisition of thermotolerance. HSP27 gene is mapped to 7q11.23. Mutant small heat-shock protein 27 causes axonal Charcot-Marie-Tooth disease and distal hereditary motor neuropathy. Heat shock protein 27 prevents cellular polyglutamine toxicity and suppresses the increase of reactive oxygen species caused by huntingtin.
